A method for tuning the selectivity of nucleic acid hybridization assays using nucleic acid biosensors and microarrays through the control of oligonucleotide immobilization density is described. The density of oligonucleotide immobilization determines the charge density at the surface by means of ionizable phosphate groups, and may result in an interfacial dielectric constant, pH and ionic strength that are unlike those of bulk solution. The density of immobilization affects the extent of interactions between neighbouring oligomers, as well as interactions between the immobilized oligomers and the substrate surface. Consequently, the thermodynamic properties of interfacial nucleic acid hybridization are substantially different from those observed in experiments done in bulk solution. The thermodynamic selectivity of a hybridization assay with respect to single base-pair mismatches (SBPMs) can be selected and enhanced using immobilized nucleic acids at particular densities.
