A polypeptide self-antigen useful in a tumor-specific vaccine mimics one or more epitopes of an antigen uniquely expressed by cells of the tumor. The polypeptide is preferably produced in a plant that has been transformed or transfected with nucleic acid encoding the polypeptide and is obtainable from the plant in correctly folded, preferably soluble form without a need for denaturation and renaturation. This plant-produced polypeptide is immunogenic without a need for exogenous adjuvants or other immunostimulatory materials. The polypeptide is preferably an scFv molecule that bears the idiotype of the surface immunoglobulin of a non-Hodgkin's (or B cell) lymphoma. Upon administration to a subject with lymphoma, the plant-produced, tumor-unique scFv polypeptide induces an idiotype-specific antibody or cell-mediated immune response against the lymphoma.
Cette invention se rapporte à un autoantigène de polypeptide, utile dans un vaccin spécifique selon les tumeurs, qui imite un ou plusieurs épitopes d'un antigène uniquement exprimé par des cellules de la tumeur. Ce polypeptide est de préférence produit dans une plante ayant été transformée ou transfectée avec de l'acide nucléique codant ledit polypeptide, lequel peut être obtenu à partir de la plante sous forme correctement repliée et de préférence soluble, sans qu'il soit nécessaire de recourir à une dénaturation ou à une renaturation. Ce polypeptide produit à partir d'une plante est immunogène sans qu'il soit nécessaire de recourir à des adjuvants exogènes ou à d'autres substances immunostimulatrices. Ce polypeptide est de préférence une molécule scFv qui porte l'idiotype de l'immunoglobuline de surface d'un lymphome d'un type autre que Hodgkin (ou lymphome B). Lorsqu'il est administré à un sujet souffrant de lymphomes, ce polypeptide scFv unique à la tumeur, produit à partir d'une plante, induit un anticorps spécifique de l'idiotype ou une réaction immunitaire à médiation cellulaire contre le lymphome.
