The present invention is directed to methods for resisting or promoting template independent nucleotide addition to the 3' terminus of a DNA duplex. The process comprises amplifying a target nucleic acid using primers which comprise a 5' terminal sequence which resists or promotes non-templated nucleotide addition to the 3' terminus of the complementary nucleic acid strand. The invention is also directed to a kit for cloning 3' nucleotidylated duplex DNA.

La présente invention se rapporte à des procédés permettant de résister à ou de favoriser l'addition nucléotidique non matricée au niveau de l'extrémité 3' d'une double hélice d'ADN. Ledit procédé consiste à amplifier un acide nucléique cible au moyen d'amorces qui comportent une séquence terminale 5' qui résiste à ou favorise l'addition nucléotidique non matricée au niveau de l'extrémité 3' du brin d'acide nucléique complémentaire. L'invention se rapporte également à un kit conçu pour le clonage d'ADN double hélice portant un groupe nucléotidyle en 3'.
