The invention relates to new compounds for cancer therapy or diagnosis and more specifically to the use of a non-toxic B subunit of Shiga toxin mutant as a vector for diagnostic products or drugs in over-expressing Gb3 receptor cells, such compounds having the following formula: STxB-Z(n)-Cys-Y(m)- T wherein - STxB is the Shiga Toxin B subunit or a functional equivalent thereof, - Z(n) wehrein n is 0 or 1 and when n is 1, Z is an amino-acid residue devoidof sulfydryl group, or is a polypeptide, - Cys is the amino-acid residue for Cysteine, - T is a molecule linked by a covalent bound to the S part of Cys, selected in a group comprising: . agents for in vivo diagnosis, . cytotoxic agents, . prodrugs, or . enzymes for the conversion of a prodrug to a drug, - Y(m) wherein m is 0 or 1 and when m is 1, Y is a linker between T and Cys, said linker being either cleavable or not cleavable for the release of T after the internalization of the hybrid compound into said cells.
L'invention concerne de nouveaux composés pour le traitement et le diagnostic du cancer et, plus particulièrement, l'utilisation d'une sous-unité B non toxique d'un mutant de toxine Shiga, en tant que vecteur pour des produits de diagnostic ou des médicaments dans une sur-expression de cellules de récepteurs Gb¿3?, de tels composés ayant la formule ci-après: STxB-Z(n)-Cys-Y(m)- T où STxB désigne la sous-unité B de toxine Shiga ou un équivalent fonctionnel de celle-ci ; Z(n) où n est égal à 0 ou 1 et, lorsque n est égal à 1, Z désigne un reste aminocacide dépourvu de groupe sulfhydrile, ou désigne un polypeptide ; Cys désigne le reste aminoacide de la cystéine ; T désigne une molécule liée, par liaison covalente, à la partie S de Cys, choisie dans un groupe comprenant : des agents de diagnostic <i>in vivo</i>, des agents cytotoxiques, des promédicaments, ou des enzymes pour la conversion d'un promédicament en un médicament ; Y(m), où m est égal à 0 ou 1 et, lorsque m est égal à 1, Y désigne un segment de liaison entre T et Cys, ledit segment de liaison étant clivable ou non-clivable pour la libération de T, après internalisation du composé hybride dans lesdites cellules.
