The present invention relates to a polymyxin derivative wherein the derivative has a total of three positive charges at physiological pH and wherein the terminal moiety (D) of the derivative comprises a total of 1 to 5 carbon atoms; and to a com-bination product comprising at least two such derivatives. The invention further relates to a method for sensitizing Gram-negative bacteria to an antibacterial agent by administering, simultaneously or sequentially in any order a therapeutically effective amount of said antibacterial agent and a derivative according to the present invention to said subject; to methods for developing novel an-tibiotics; and for sensitizing clinically important bacteria to a host defense mechanism complement present in serum. The inven-tion also relates to a method of treating a subject for a gram-negative bacterial infection by administering a polymyxin derivative of the invention in combination with a second antibacterial agent. Finally, the invention relates to a process for preparing such polymyxin derivatives.
La présente invention concerne un dérivé de polymyxine, le dérivé ayant un total de trois charges positives à un pH physiologique et la fraction terminale (D) du dérivé comprenant un total de 1 à 5 atomes de carbone; et un produit mixte comprenant au moins deux de ces dérivés. L'invention concerne en outre un procédé pour sensibiliser des bactéries à gram négatif à un agent antibactérien par ladministration, simultanément ou séquentiellement dans nimporte quel ordre, dune quantité thérapeutiquement efficace dudit agent antibactérien et dun dérivé selon la présente invention audit sujet; des procédés pour développer de nouveaux antibiotiques; et pour sensibiliser des bactéries cliniquement importantes à un complément de mécanisme de défense de l'hôte présent dans le sérum. L'invention concerne également une méthode de traitement dun sujet pour une infection bactérienne à gram négatif par ladministration dun dérivé de polymyxine de linvention en association avec un second agent antibactérien. Enfin, linvention concerne un procédé pour préparer de tels dérivés de polymyxine.
