The current invention comprises a method for producing a heterologous polypeptide with a reduced degree of fucose modification in a mammalian cell by cultivating the mammalian cell under conditions suitable for the expression of said heterologous polypeptide, and recovering the heterologous polypeptide from the mammalian cell or the culture, wherein in said mammalian cell the enzymatic activity of .alpha.l,6-fucosyltransferase is reduced by means of an shRNA directed against .alpha.l,6-fucosyltransferase mRNA.
La présente invention concerne un procédé de production d'un polypeptide hétérologue avec un degré réduit de modification fucosidique dans une cellule de mammifère. Ledit procédé consiste à cultiver la cellule de mammifère dans des conditions appropriées pour l'expression dudit polypeptide hétérologue, et à récupérer le polypeptide hétérologue à partir de la cellule de mammifère ou de la culture, l'activité enzymatique de l'.alpha.-l,6-fucosyltransférase dans ladite cellule de mammifère étant réduite au moyen d'un ARNsh dirigé contre l'ARNm de l'.alpha.-l,6-fucosyltransférase.
