The present invention provides a method of transducing the conformational change undergone by a signaling aptamer upon binding a ligand to a differential signal generated by a reporter molecule. Also provided is a method of detecting and quantitating a ligand in solution using an aptamer conjugated to a fluorescent dye (signaling aptamer) to bind to the ligand and measuring the resultant optical signal generated.
La présente invention concerne une méthode permettant de transduire le changement conformationnel subi par un aptamère de signalisation lors de sa liaison à un ligand en un signal différentiel généré par une molécule reporter. L'invention concerne également une méthode de détection et de quantification d'un ligand dans une solution, utilisant un aptamère conjugué à un colorant fluorescent (aptamère de signalisation) pour la liaison avec le ligand, et une méthode de mesure du signal optique généré obtenu.
