A single chain T cell receptor (scTCR) comprising an a segment constituted by a TCR .alpha. chain variable region sequence fused to the N terminus of a TCR .alpha. chain constant region extracellular sequence, a .beta. segment constituted by a TCR .beta. chain variable region fused to the N terminus of a TCR .beta. chain constant region extracellular sequence, and a linker sequence linking the C terminus of the .alpha. segment to the N terminus of the .beta. segment, or vice versa, the constant region extracellular sequences of the .alpha.and .beta. segments being linked by a disulfide bond, the length of the linker sequence and the position of the disulfide bond being such that the variable region sequences of the .alpha. and .beta. segments are mutually orientated substantially as in native .alpha..beta. T cell receptors. Complexes of two or more such scTCRs, and use of the scTCRs in therapy and in various screening applications are also disclosed.
L'invention concerne un récepteur des lymphocytes T monocaténaire (scTCR) comprenant un segment constitué par une séquence de région variable à chaîne .alpha. TCR fondue au N-terminal d'une séquence extracellulaire de région constante à chaîne .alpha. TCR, un segment .beta. constitué d'une région variable à chaîne .beta. TCR fondu à une séquence extracellulaire de région constante à chaîne .beta. TCR, et une séquence de liaison du C-terminal du segment .alpha. au N-terminal du segment .beta., ou vice versa, les séquences extracellulaires de région constante des segments .alpha. et .beta. étant liées par une liaison disulfide, la longueur de la séquence de liaison et la position de la liaison disulfide étant telles que les séquences de région variable des segments .alpha. et .beta. sont mutuellement orientées sensiblement comme dans les récepteurs de lymphocytes T .alpha. et .beta. natifs. L'invention concerne également des complexes de deux ou plusieurs scTCRs de ce type, ainsi que l'utilisation des scTCRs en thérapie et dans diverses applications de criblage.
