ABSTRACT The invention described is a test method and testing kit used in the detection and quantitation of complement fixation antimicrobial antibodies, such as antiviral antibodies, in human or animal serum samples. The test procedure utilizes Staphylococcal protein A-coated red blood cells immobilized in a gel into which is encorporated a specific microbial antigen. Undiluted heat inactivated serum samples are allowed to diffuse radially from wells punched in the solidified gel. Protein A serves to concentrate the subsequent antigen-antibody reaction on the surface of the red blood cell. Zones of hemolysis are developed by flooding with complement. The diameter Or the zone of hemolysis is proportional to the concentration of specific antibody in the serum. Using adenovirus as a model, basic test parameters have been defined and optimum reagent concentrations and diffusion conditions have been determined.
