The authors describe an expression system for the correct, stable and effective expression in mammalian cells of a siRNA comprising: a) a polymerase II RNA-dependent promoter derived from the U1 RNA gene; downstream from the sequence b) suitable restriction sites for the cloning of the sequence transcribing the pre-siRNA; downstream from these sites c) sequences derived from the sequences at 3~ of the gene for U1 snRNA that are necessary and sufficient for the correct formation of 3~ of the presiRNA.

L'invention concerne un système d'expression permettant l'expression correcte, stable et efficace d'un ARNsi dans des cellules de mammifère. Ce système comprend : a) un promoteur dépendant de l'ARN polymérase II, obtenu à partir du gène d'ARN U1 ; en aval de la séquence, b) des sites de restriction convenant pour le clonage de la séquence transcrivant le pré-ARNsi ; en aval de ces sites, c) des séquences obtenues à partir des séquences se situant en position 3' du gène d'ARNsn U1, qui sont nécessaires et suffisantes pour la formation correcte en position 3' du pré-ARNsi.
