Mono-pegylated arginase conjugate and method producing thereof. The mono- pegylated arginase is homogeneous in molecular weight and shows therapeutic effect for treating cancers and viral infections. The method of producing such arginase conjugate has a main step of genetically modifying the gene encoding an arginase so that the PEG moiety can attach to the enzyme at a predetermined, specific intended site. This is achieved by removing the PEG attaching amino acid residues at undesirable sites while keeping (or adding, if necessary) the one at the desirable site of the enzyme. Two exemplary mono-pegylated arginase con-jugates so produced are human arginase I (HAI) where a polyethylene glycol (PEG) moiety is site-specific covalently bonded to Cys45 of the enzyme and Bacillus caldovelox arginase (BCA) where a polyethylene glycol (PEG) moiety is site-specific covalently bonded to Cys161 of the enzyme.
L'invention porte sur un conjugué d'arginase monopégylée et sur son procédé de production. L'arginase monopégylée est homogène en poids moléculaire et montre un effet thérapeutique pour traiter des cancers et des infections virales. Le procédé de production d'un tel conjugué d'arginase a une étape principale de modification génétique du gène codant pour une arginase de telle sorte que la fraction PEG peut se fixer à l'enzyme au niveau d'un site destiné spécifique, prédéterminé. Ceci est réalisé en enlevant les résidus d'acides aminés de fixation à PEG sur les sites indésirables tout en gardant (ou en ajoutant, si nécessaire) celui situé sur le site désirable de l'enzyme. Deux conjugués d'arginase monopégylée à titre d'exemples produits de cette façon sont l'arginase I humaine (HAI) dans laquelle une fraction polyéthylène glycol (PEG) est liée de façon covalente et spécifique de site à Cys45 de l'enzyme et l'arginase de Bacillus caldovelox (BCA) dans laquelle une fraction polyéthylène glycol (PEG) est liée de façon covalente et spécifique de site à Cys161 de l'enzyme.
