A method is described for the site-specific preparation of hGRF-PHG conjugates containing one or more PEG units (per mole of hGRF) covalently bound to Lys12 and/or Lys21 and/or N.alpha., characterized in that the conjugation reaction between the hGRF peptide and activated PEG is carried out in solution and the desired hGRF-PEG conjugate can be purified by chromatography. The conjugates prepared by this method, as well as their use in the treatment, prevention or diagnosis of growth hormone deficiency, are also an object of the present invention.

L'invention concerne un procédé de préparation spécifique à un site de conjugués de hGRF-PEG contenant un ou plusieurs unités de PEG (par mole de hGRF) liées par covalence à Lys<12> et/ou Lys<21> et/ou N< alpha >, procédé caractérisé en ce que la réaction de conjugaison entre le peptide hGRF et le polyéthylène-glycol activé s'effectue en solution et que le conjugué de hGRF-PEG voulu peut être purifié par chromatographie. L'invention concerne également les conjugués préparés conformément au procédé de l'invention, ainsi que leur utilisation dans le traitement, la prévention et le diagnostic d'un défaut de croissance hormonale.
