A method for detecting multiple SNPs (singe nucleotide polymorphisms) in a population of target polynucleotides comprising combining target polynucleotides, capture polynucleotides and SNP probes wherein each target comprises a different capture region and a different SNP region, the capture polynucleotides are immobilized and arrayed at corresponding discrete elements on a substrate and each capture polynucleotide comprises a sequence which specifically hybridizes to a corresponding different capture region, each SNP probe comprises a sequence complementary to a corresponding different SNP region, and the target polynucleotides are immobilized by hybridizing to the capture polynucleotides; and detecting the presence of each SNP probe on the substrate wherein the presence of a given SNP probe at a given element indicates the presence of the corresponding SNP in the corresponding target polynucleotide.

Cette invention concerne des procédés, des compositions et des systèmes de détection de multiples polymorphismes nucléotidiques simples (SNP) dans une population de polynucléotides cibles en parallèle avec une analyse biologique d'alternance d'échantillons SNP, de polynucléotides de capture et, éventuellement, de polynucléotides auxiliaires. Les affinités relatives des échantillons SNP pour les zones SNP correspondantes peuvent être augmentées au moyen de réactifs qui régularisent les températures de fusion des échantillons, et/ou en facilitant localement les interactions entre l'échantillon SNP, la zone SNP, le polynucléotide de capture et/ou les polynucléotides auxiliaires, par exemple au moyen d'un liant d'association à la rainure mineure. Les échantillons peuvent comprendre un ensemble dégénéré regroupant tous les polynucléotides possibles de même taille et les polynucléotides de capture sont en général immobilisés et classés par ordre au niveau des éléments discrets correspondants à haute densité.
