The present invention provides methods, apparatus and kits for synthesizing assembled peptides and proteins on a solid phase with sequential ligation of three or more unprotected peptide segments using chemoselective and mild ligation chemistries in aqueous solution. Also provided are methods of monitoring solid phase sequential ligation reactions using MALDI or electrospray ionization mass spectrometry of reaction products.

La présente invention concerne des procédés, des appareils et des trousses permettant de synthétiser des peptides et des protéines assemblés sur une phase solide, avec une ligation séquentielle de trois ou davantage de segments de peptides non protégés mise en oeuvre au moyen de produits chimiques de ligation modérés et chimiosélectifs dans une solution aqueuse. L'invention concerne également des procédés permettant de surveiller des réactions de ligation séquentielle en phase solide à l'aide d'une spectrométrie de masse de type MALDI ou d'une spectrométrie de masse d'ionisation par pulvérisation d'électrons effectuée sur des produits de réaction.
