The invention includes deleting codon segments from DNA expressing a native protein in order to obtain a shorter, soluble protein which mimics characteristics of an insoluble native protein. Soluble proteins of the invention are characterized by: (1) having less amino acids than the full length native protein; (2) having a higher degree of solubility than the native protein; (3) retaining the basic biological characteristics of the native protein such as (a) not being subject to enzymatic digestion and (b) causing disease. Soluble proteins of the invention are obtained by providing a DNA sequence which encodes a native protein and systematically removing codons, making copies of the shortened versions of DNA which are then expressed to provide the shortened proteins. The shortened proteins are then tested for solubility. Soluble proteins are then further tested to confirm that they retain the biological characteristics of the native protein.

L'invention consiste à supprimer des segments de codon d'un ADN exprimant une protéine native, en vue d'obtenir une protéine soluble, plus courte, présentant des caractéristiques analogues à celles d'une protéine native insoluble. Les protéines solubles de l'invention sont caractérisées: (1) en ce qu'elles possèdent moins d'acides aminés que les protéines natives entières; (2) en ce qu'elles possèdent un degré de solubilité plus élevé que les protéines natives; (3) et en ce qu'elles conservent les caractéristiques biologiques de base des protéines natives, c'est-à-dire (a) qu'elles ne sont pas sujettes à la digestion enzymatique et (b) qu'elles causent des maladies. Pour obtenir les protéines solubles de l'invention, on prend une séquence d'ADN codant pour une protéine native et on en supprime systématiquement les codons, on fait des copies des versions raccourcies d'ADN, qui sont ensuite exprimées en vue d'obtenir les protéines raccourcies. On teste, ensuite, la solubilité des protéines raccourcies. Puis, on teste les protéines solubles afin de vérifier si elles conservent les caractéristiques biologiques de la protéine native.
