The invention relates to a soluble Rubella E1 antigen and variants lacking at the C--terminal end at least the transmembran region and the anchor segment as well as at least the amino acids 143 to 164 and containing at least the region spanning the disulfid-bridges Cys 349 - Cys 352 and Cys 368 - 401 whereas the N-terminus of this region contains additionally at least 15 amino acids and/or the C- terminus of this region contains additionally at least 8 amino acids of the adjacent Rubella E1 antigen sequence. The region spanning the disulfid-bridges Cys 349 - Cys 352 and Cys 368 - 401 contains at the N-terminus of this region additionally at least 25, 30, 34 amino acids and/or at the C-terminus of this region additionally at least 10, 11, 15, 25, 35 amino acids of the adjacent Rubella E1 antigen sequence. Furthermore, the invention relates to a recombinant DNA molecule encoding a Rubella E1 antigen and variants, methods for the detection, determination and quantification of anti-Rubella antibodies, and a diagnostic test and reagent kit for the detection of anti-Rubella antibodies.
L'invention porte sur un antigène soluble E1 du virus de la rubéole et de variants ne présentant pas à l'extrémité C-terminale au moins la région transmembranaire et le segment d'ancrage, de même qu'au moins les acides aminés 143 à 164 et contenant au moins la région couvrant les ponts disulfure Cys 349 à Cys 352 et Cys 368 à 401, où l'extrémité N-terminale de cette région contient au moins 15 autres acides aminés et/ou l'extrémité C- terminale de cette région contient au moins 8 autres acides aminés de la séquence adjacente de l'antigène E1 du virus de la rubéole. La région couvrant les ponts disulfure Cys 349 à Cys 352 et Cys 368 à 401 contient à l'extrémité N-terminale de cette région au moins 25, 30, 34 autres acides aminés et/ou à l'extrémité C-terminale de cette région au moins 10, 11, 15, 25, 35 autres acides aminés de la séquence adjacente de l'antigène E1 du virus de la rubéole. En outre, l'invention concerne une molécule d'ADN recombinante codant un antigène E1 du virus de la rubéole et des variants, des méthodes permettant la détection, la caractérisation et la quantification d'anticorps anti-rubéole et une épreuve diagnostique ainsi qu'une trousse de réactifs destinés à la détection d'anticorps anti-rubéole.
