New solid drug dispersions are described in which the drug is present in amorphous form and massively dispersed (in bulk) inside the particles of an organic carrier selected from cross-linked polymers and/or a complexing agents. These dispersions are obtainable by mixing together the drug and the carrier and applying an oscillating electromagnetic field to the mixture, to a frequency belonging to the microwave region; the microwaves are applied according to a specific heating cycle wherein the drug-carrier mixture is heated at a temperature higher than the melting point of the drug for at least 5 minutes. With respect to the known techniques, the present invention allows to increase in the amount of drug incorporated into the carrier in amorphous form, and to increase the physical stability of the amorphous phase. This is particularly useful in the preparation of pharmaceutical compositions based on drugs which are crystalline in nature, such as are notoriously sparingly soluble in water: thanks to the increased amounts and stability of the drug in amorphous form, the resulting formulations have a more rapid and intense effect, and are endowed with greater bioavailability.
L'invention concerne de nouvelles dispersions de médicaments solides, dans lesquelles le médicament est présent sous forme amorphe et massivement dispersé (en vrac) à l'intérieur des particules d'un support organique sélectionné parmi des polymères réticulés et/ou d'agents de complexage. On peut obtenir ces dispersions en mélangeant le médicament et le support et en appliquant au mélange un champ électromagnétique oscillant, à une fréquence appartenant à la zone d'hyperfréquences. Ces hyperfréquences sont appliquées en fonction d'un cycle de chauffage spécifique, ledit mélange de support de médicaments étant chauffé à une température supérieure au point de fusion du médicament pendant au moins 5 minutes. Par rapport aux techniques connues, ledit procédé de l'invention permet d'augmenter la quantité de médicaments incorporés dans le support sous sa forme amorphe et d'accroître la stabilité physique de la phase amorphe. Ceci est particulièrement utile dans la préparation de compositions pharmaceutiques reposant sur des médicaments qui sont cristallins de nature, notoirement modérément solubles dans l'eau, en raison des quantités et de la stabilité accrues du médicament sous sa forme amorphe. Les préparations résultantes ont alors un effet plus rapide et intense et elles présentent une biodisponibilité supérieure.
