Robert Paul Cory STABILIZED GLUCOSE ?MERASE ENZYME CONCENTRATE AND PROCESS FOR PREPARING THE SAME ABSTRACT OF THE DISCLOSURE A stabilized glucose isomerase enzyme concentrate comprising a substantially water soluble and nucleic acid-free enzyme con- centrate of (1) a cell-free glucose isomerase enzyme and (2) a substantially water soluble magnesium salt characterized as having: (a) a protein content ranging from about 50 to about 80%, by weight, dry basis; (b) a Mg++ content ranging from about 3 to about 45 mg. of Mg++ per ml. of enzyme concentrate; (c) a Mg++/protein ratio ranging from about 0.02 to about 0.75; (d) a specific isomerase activity of at least about 10 IGIU/mg of protein; and (e) a stability such that it retains at least about 95% of its initial isomerase activity when stored at 26°C. for up to 30 days and 80?10% of its initial isomerase activity when stored at 18°C. for up to one year. The enzyme concentrate will generally have an isomerase activity of at least 5,000 IGIU/gram, dry basis and preferably at least 8,000 IGIU/gram, dry basis, and contain a water miscible organic solvent such as 2-propanol. The stabilized enzyme concentrate is prepared by contacting an aqueous mixture containing cell-free glucose isomerase and a water miscible organic solvent with a small, but effective amount of a substantially water soluble magnesium salt. This treatment causes the formation of an enzyme-magnesium precipitate. The precipitate (the stabilized enzyme concentrate) can be recovered by conventional techniques such as by centrifugation. The stabilized enzyme concentrate can be used to place a high level of enzyme per unit volume on most water insoluble carriers (e.g., DEAE cellulose; synthetic anion exchange resins; porous, water insoluble materials such as basic magnesium carbonate and porous alumina materials) to form a biocatalyst useful to continuously convert glucose to fructose.
