The invention relates to a recombinant influenza virus for high-yield expression of incorporated foreign gene(s), which is genetically stable in the absence of any helper virus and which comprises at least one viral RNA segment being an ambisense RNA molecule (ambisense RNA segment) and containing one of the standard viral genes in sense orientation and a foreign, recombinant gene in anti-sense orientation, or vice versa, in overall convergent arrangement. The present invention further provides a method for the production of said recombinant influenza virus, a method for constructing specific ribozyme- sensitive influenza carrier strains; pharmaceutical compositions comprising said recombinant influenza virus; and the use of said recombinant influenza virus for preparing medicaments for vaccination purposes.

L'invention concerne un virus de recombinaison de la grippe permettant l'expression d'un rendement élevé de gènes étrangers incorporés, génétiquement stable en l'absence de virus assistant, comprenant au moins un segment d'ARN viral représenté par une molécule d'ARN à deux sens (segment d'ARN à deux sens), et contenant l'un des gènes viraux standards dans une orientation sens et un gène de recombinaison étranger dans un orientation antisens ou vice-versa, dans un agencement convergeant global. L'invention concerne également une technique de production dudit virus de recombinaison de la grippe, et une technique de construction de souches porteuses spécifiques de grippe sensibles à un ribozyme. Elle concerne enfin des compositions pharmaceutiques comprenant ledit virus de recombinaison de la grippe et l'utilisation de ce virus pour préparer des médicaments à des fins de vaccination.
