The present invention relates to a vector system for use in the generation of nucleic-acid libraries and in methods of cosuppression. The invention resides in the use of a vector comprising a single-stranded loop portion and a single-stranded replicon portion separated by a double-stranded portion comprising at least one unique restriction endonuclease site.
