The present invention relates to various methods of genetic immunization for the purpose of providing antigen-specific immunity in a mammalian host, including a human host. The invention is based on the ability to direct particulate polynucleotides which express an antigenic protein or protein fragment to the cytoplasm of host target cells, such as antigen presenting cells. A directed delivery of such particulate polynucleotides to the cytoplasm of antigen presenting cells will stimulate antigen-specific CTL production, thus promoting destruction of affected cells such as neoplastic cells and virally infected cells.
L'invention concerne différentes méthodes d'immunisation génétique permettant de conférer une immunité à un mammifère hôte vis-à-vis d'un antigène spécifique, en particulier un hôte humain. L'invention se base sur la faculté de diriger des polynucléotides particulaires, capables d'exprimer une protéine ou un fragment de protéine antigénique, sur le cytoplasme des cellules cibles hôtes, telles que les cellules présentant l'antigène. Un apport direct de ces polynucléotides particulaires dans le cytoplasme des cellules présentant l'antigène va stimuler la production de lymphocytes cytotoxiques spécifiques de l'antigène, ce qui va favoriser la destruction de cellules malades, telles que les cellules néoplasiques et les cellules infectées par un virus.
