A method for preparing human dendritic cells containing antigens by pulsing human dendritic cells with cationic liposomes comprising at least one antigen to obtain antigen pulsed human dendritic cells is disclosed. Additionally, a method of producing antigen specific human CD8+ cytotoxic T-lymphocytes (CTL) and CD4+ helper T-lymphocytes by incubating human T-lymphocytes with the antigen pulsed human dendritic cells produced by the disclosed method under conditions that promote the generation of antigen-specific T-lymphocytes with specificity to the antigen with which the dendritic cells were pulsed is disclosed. Use of the antigen pulsed dendritic cells in immunotherapy and for prophylactic vaccination also is disclosed. Additionally, use of the stimulated antigen specific human CD8+ cytotoxic T-lymphocytes (CTL) and CD4+ helper T-lymphocytes in adoptive immunotherapy and for prophylactic vaccination is also disclosed. Compositions containing antigen pulsed human dendritic cells and/or antigen specific human CD8+ cytotoxic T-lymphocytes (CTL) and CD4+ helper T-lymphocytes produced by the methods of the present invention are also disclosed. The methods and compositions are particularly useful for stimulating the immune response against human papillomavirus (HPV) oncoproteins full-length E7 or E6 that have been associated with human cervical cancer.
