A strategically modified hepatitis B core protein is described, where an insert is provided, preferably in an immunodominant region of the nucleocapsid protein, containing a chemically reactive amino acid residue. The modified hepatitis B core protein or its aggregated nucleocapsid protein particles can be pendently linked to a hapten to form a modified nucleocapsid conjugate. Such a conjugate is useful in the preparation of vaccines or antibodies. The modified hepatitis B core protein can also be modified to include a T cell epitope.
L'invention concerne une protéine noyau d'hépatite B stratégiquement modifiée, avec insert, de préférence dans une région immunodominante de la nucléocapside, renfermant un résidu d'acide aminé chimiquement réactif. Il est possible de réaliser une liaison pendante entre la protéine noyau d'hépatite B modifiée ou des particules agrégées de nucléocapside, de manière à former un conjugué de nucléocapside modifié. Ce type de conjugué est utile pour l'élaboration de vaccins ou d'anticorps. On peut aussi modifier la protéine noyau d'hépatite B modifiée pour y incorporer un épitope de cellule T.
