The present invention relates to an agent for reducing ischaemic damage to an organ, in particular to a heart and a brain, pharmaceutical compositions comprising said agent and the use of said agent for the treatment of ischaemic diseases to the heart and the brain. The agent comprises a substituted 4- phenyl-4-[1H-imidazol-2-yl]-piperidine derivative according to Formula (I) the pharmaceutically acceptable acid or base addition salts thereof, the stereochemically isomeric forms thereof, the tautomeric forms thereof, the N- oxide forms thereof and the prodrugs thereof. In particular are claimed the compounds according to Formula (I) in which A=B is C=O or SO2, X is a covalent bond, R1 is alkyloxy, alkyloxyalkyl, Ar or NR9R10, wherein R9 and R10 each independently are hydrogen or Ar ; or A=B and R1 together form a benzoxazolyl radical ; p is zero, R3 is benzyl optionally substituted with hydroxy, alkyl or alkyloxycarbonyl and R4 and R5 each are hydrogen. The use of said agents has important clinical ramifications with regard to the reduction of ischaemic damage to an organ in a mammal, in particular to a heart and/or a brain, the prevention of coronary artery diseases in a mammal by inducing a cardioprotective effect and the treatment and prevention of stroke.
La présente invention concerne un agent qui permet de réduire les lésions ischémiques d'un organe, notamment le coeur et le cerveau, des compositions pharmaceutiques renfermant cet agent et l'utilisation dudit agent pour traiter les maladies ischémiques du coeur et de l'encéphale. L'agent comprend un dérivé de 4-phényl-4-[1H-imidazol-2-yl]-pipéridine substituée représenté par la formule (I). Cette invention concerne cet agent ainsi que les sels d'addition d'acide ou de base de ce dernier, les formes stéréochimiquement isomères de ce dernier, les formes tautomères de ce dernier, les formes N-oxyde de ce dernier et les promédicaments de ce dernier. Cette invention se rapporte plus particulièrement aux composés représentés par la formule (I) dans laquelle A=B et C=O ou SO¿2?, X représente une liaison covalente, R?1¿ représente alkyloxy, alkyloxalkyle, Ar ou NR?9¿R?10¿, où R?9¿ et R?10¿ représentent chacun indépendamment hydrogène ou Ar; ou A=B et R?1 ¿forment ensemble un radical benzoxazolyle; p représente zéro, R?3¿ représente benzyle éventuellement substitué par hydroxy, alkyle ou alkyloxycarbonyle et R?4¿ et R?5¿ représentent chacun hydrogène. L'utilisation de ces agents présente des implications cliniques importantes en qui concerne la réduction des lésions ischémiques d'un organe d'un mammifère, notamment le coeur ou l'encéphale, la prévention des coronaropathies chez un mammifère par induction d'un effet cardioprotecteur, et le traitement et la prévention des accidents vasculaires cérébraux.
