Compounds of formula (I) where R1 is C1-C4 alkyl; R2 is H, -OR1, -SR1, - NH(R1), -N(R4)(R5), aminocarbonyl, halogen, or -CN, where R4 and R5 are independently C1-C6 alkyl, or together form a 3- to 7- membered heterocycloalkyl, such heteroatom selected from oxygen, nitrogen, and sulfur, optionally including an additional heteroatom which is nitrogen, oxygen, or sulfur, R3 is cyclopentyl or (a) where R' and R" are independently H=O, -OH, or -NH2 or R' and R" can together form (b), R"' is -OH or -NH2; provided that when R3 is cylopentyl or norbonyl, R2 cannot be H or halogen; and pharmaceutically-acceptable salts thereof. These are diuretics; renal protectives against renal failure; agents to facilitate recovery from coma, to improve the therapeutic outcome resulting from defibrillation or cardiopulmonary resuscitation by preventing post-resuscitation bradycardia, bradyarrhythmia and cardioplegia, to restore cardiac function following a cardioplegia procedure, and to treat or prevent intermittent claudication.

Composés de la formule (I), dans laquelle R1 représente alkyle C1-C4; R2 représente H, -OR1, -SR1, -NH(R1), -N(R4)(R5), aminocarbonyle, halogène ou -CN, R4 et R5 représentant indépendamment alkyle C1-C6, ou formant ensemble un hétérocycloalkyle de 3 à 7 chaînons, un hétéroatome tel que choisi entre oxygène, azote et soufre, et comprenant éventuellement un hétéroatome supplémentaire tel qu'azote, oxygène ou soufre; R3 représente cyclopentyle ou (a), R' et R" représentant indépendamment H=O, -OH ou -NH2, ou R' et R" formant ensemble la structure (b) en tant que R3, R"' représentant -OH ou -NH2; sachant que, lorsque R3 représente cyclopentyle ou norbornyle, R2 ne peut représenter H ou halogène; et leurs sels pharmaceutiquement acceptables. Ces composés sont des diurétiques; des agents de protection rénale contre l'insuffisance rénale; des agents favorisant le rétablissement suite à un coma, et améliorant les résultats thérapeutiques d'une défibrillation ou d'une réanimation cardio-pulmonaire en prévenant la bradycardie, la bradyarythmie et la cardioplégie survenant après une telle réanimation; des agents favorisant le rétablissement de la fonction cardiaque suite à une intervention relative à la cardioplégie; ainsi que des agents permettant de traiter ou de prévenir une claudication intermittente.
