The present invention relates to compounds of formula (I) wherein R1 is - (CRR')n-aryl-(CRR')n-heteroaryl, -(CRR')n-O-aryl or -(CRR')n-O-heteroaryl, wherein the aryl or heteroaryl groups are unsubstituted or substituted by one or two substituents, selected from the group consisting of halogen, lower alkyl, lower alkoxy, lower alkyl substituted by halogen, or unsubstituted or substituted phenyl, wherein the substitution on the phenyl group maybe lower alkyl or halogen, or is -(CRR')n-2-benzo[1.3]dioxolyl; R/R'are independently from each other hydrogen, lower alkyl or are together with the carbon atom to which they are attached a cycloalkyl group; R2/R3 are independently from each other hydrogen, lower alkyl, phenyl or cycloalkyl or may form together with the N- atom to which they are attached a five or six membered saturated heterocycle; n is O or 1; and to pharmaceutically suitable acid addition salts thereof. The invention includes all sterioisomeric forms, including individual diastereoisomers and enantiomers of the compound of formula (I) as well as racemic and non-racemic mixtures thereof. It has been found that the present compounds are selective allosteric positive modulators of the human NK-3 receptor for the treatment of anxiety, depression, bipolar disorders, Parkinson's disease, schizophrenia and pain.

La présente invention concerne des composés de formule (I) dans laquelle R1 est un groupe -(CRR')n-aryle, -(CRR')n-hétéroaryle, -(CRR')n-O-aryle ou -(CRR')n-O-hétéroaryle, les groupes aryles ou hétéroaryles étant non substitués ou substitués par un ou deux substituants sélectionnés chacun entre un halogène, un alkyle inférieur, un alcoxy inférieur, un alkyle inférieur substitué par un halogène ou un phényle substitué ou non substitué, la substitution sur le groupe phényle pouvant être un alkyle inférieur ou un halogène, ou est un groupe -(CRR')n-2-benzo[1.3]dioxolyle ; R/R' sont chacun indépendamment de l'autre un hydrogène, un alkyle inférieur ou bien ils forment ensemble avec l'atome de carbone auquel ils sont attachés un groupe cycloalkyle ; R2/R3 sont chacun indépendamment de l'autre un hydrogène, un alkyle inférieur, un phényle ou un cycloalkyle ou bien ils peuvent former avec l'atome de N auquel ils sont attachés un hétérocycle à cinq ou six chaînons saturé ; n est 0 ou 1 ; et des sels de ceux-ci obtenus par ajout d'acide acceptables du point de vue pharmaceutique. L'invention inclut toutes les formes stéréoisomériques, dont les diastéréoisomères et les énantiomères séparés des composés de formule (I) ainsi que les mélanges racémiques et non racémiques de ceux-ci. On a trouvé que les présents composés sont des modulateurs positifs allostériques sélectifs du récepteur NK-3 humain pour le traitement de l'anxiété, de la dépression, de psychoses maniacodépressives, de la maladie de Parkinson et de la douleur.
