Methods and compositions for amplification of a target polynucleotide sequence by suppressing amplification of related sequences are provided. The method uses a polymerase lacking 5'-3' exonuclease activity and a non-extendable blocker oligonucleotide that hybridizes preferentially to a non-target sequence and downstream of a primer, thereby preventing template--dependent primer extension by the polymerase.
La présente invention propose des procédés et des compositions pour l'amplification d'une séquence de polynucléotides cible par suppression de l'amplification des séquences associées. Le procédé utilise une polymérase dépourvue d'activité exonucléase 5'-3' et un oligonucléotide bloquant non extensible qui s'hybride préférentiellement à une séquence non cible et en aval d'une amorce, empêchant de cette façon l'extension de l'amorce dépendante de la matrice par la polymérase.
