A device, and method of making the device, capable of therapeutic treatment and/or for in vitro testing of human skin. The device may be used on skin wounds for burned, injured, or diseased skin, and provides structures and functions as in normal uninjured skin, such as barrier function, which is a definitive property of normal skin. The device contains cultured dermal and epidermal cells on a biocompatible, biodegradable reticulated matrix. All or part of the cells may be autologous, from the recipient of the cultured skin device, which advantageously eliminates concerns of tissue compatibility. The cells may also be modified genetically to provide one or more factors to facilitate healing of the engrafted skin replacement, such as an angiogenic factor to stimulate growth of blood vessels. The inventive device is easy to handle and manipulate for surgical transplant, can be made into large sheets to minimize the number of grafts required to cover a large surface area to be treated, and can be produced within the time frame to treat a burned individual requiring a skin graft.
L'invention concerne un dispositif et un procédé permettant à ce dispositif de mettre en oeuvre un traitement thérapeutique et/ou un test <i>in vitro</i> de la peau humaine. Ce dispositif peut être utilisé sur des lésions cutanées pour la peau brûlée, blessée, ou malade, et fournit des structures et des fonctions identiques à celles d'une peau non blessée normale, telle qu'une fonction barrière, qui est une propriété absolue d'une peau normale. Le dispositif contient des cellules dermiques et épidermiques cultivées sur une matrice réticulée biocompatible et biodégradable. Tout ou une partie des cellules peuvent être autologues, à partir du récipient du dispositif dans lequel la peau est cultivée, qui élimine de façon avantageuse des problèmes de compatibilité des tissus. Les cellules peuvent également être modifiées génétiquement en vue de fournir un ou plusieurs facteurs permettant de faciliter la guérison de remplacement de peau greffée, tels qu'un facteur angiogénique permettant de stimuler la croissance des vaisseaux sanguins. Le dispositif de l'invention est facile à manoeuvrer et à manipuler dans la transplantation chirurgicale, peut être fabriqué en grandes feuilles en vue de minimiser le nombre de greffes nécessaires pour recouvrir une grande zone superficielle à traiter, et peut être produit dans un temps donné afin de traiter un individu brûlé nécessitant une greffe de la peau.
