Linkage of myocardial infarction (MI) and a locus on chromosome 13q12 is disclosed. In particular, the FLAP gene within this locus is shown by genetic association analysis to be a susceptibility gene for MI and ACS, as well as stroke and PAOD. Pathway targeting for treatment and diagnostic applications in identifying those who are at risk of developing MI, ACS, stroke or PAOD, in particular are described. The invention also provides for compositions comprising a leukotriene synthesis inhibitor and a stating and methods of using these compositions to reduce C-reactive protein in a human subject at risk of MI, ACS, stroke and/or PAOD.
La présente invention a trait à la liaison de l'infarctus myocardique et un locus sur le chromosome 13q12. En particulier, il est démontré par l'analyse d'association génétique que le gène FLAP au sein de ce locus est un gène de prédisposition à l'infarctus myocardique et au syndrome coronaire aigu, ainsi qu'à l'accident vasculaire cérébral et la maladie d'occlusion artérielle périphérique. En particulier, l'invention a trait au ciblage de voies pour des applications thérapeutiques et diagnostiques dans l'identification de ceux susceptibles de développer l'infarctus myocardique et le syndrome coronaire aigu, l'accident vasculaire cérébral ou la maladie d'occlusion artérielle périphérique. L'invention a également trait à des compositions comportant un inhibiteur de la synthèse du leucotriène et des procédés d'utilisation de ces compositions pour la réduction de la protéine C réactive chez un sujet humain présentant une prédisposition à l'infarctus myocardique, au syndrome coronaire aigu, à l'accident vasculaire cérébral et/ou à la maladie d'occlusion artérielle périphérique
