A method is provided for synthesizing catechol from a biomass-derived carbon source capable of being used by a host cell having a common pathway of aromatic amino acid biosynthesis. The method comprises the steps of biocatalytically converting the carbon source to 3-dehydroshikimate in said host cell, biocatalytically converting the DHS to protocatechuate, and decarboxylating the protocatechuate to form catechol. Also provided is a heterologue E. coli transformant characterized by the expression of genes encoding transhetolase, DAHP synthase, and DHQ synthase, further characterized by the constitutive expression of structural genes encoding 3-dehydroshikimate dehydratase and protocatechuate decarboxylase.
Procédé de synthèse de catéchol à partir d'une source de carbone dérivée de la biomasse pouvant être utilisée par une cellule hôte ayant une voie commune de biosynthèse des acides aminés aromatiques. Ledit procédé consiste à convertir par biocatalyse la source de carbone en 3-déshydroshikimate (DHS) dans ladite cellule hôte, à convertir par biocatalyse le DHS en protocatéchuate, et à décarboxyler le protocatéchuate pour obtenir du catéchol. La présente invention concerne également un transformant de E. coli hétérologue caractérisé par l'expression de gènes codant la transhétolase, la synthase de 7-phosphate de l'acide 3-désoxy-D-arabino-heptulosonique (DAHP) et la 3-déshydroquinate synthase (DHQ) et caractérisé en outre par l'expression constitutive de gènes structuraux codant la 3-déshydroshikimate déshydratase et la protocatéchuate décarboxylase.
