The original synthesis of combretastatin A-2 (1a) was modified to provide an efficient scale-up procedure for obtaining this antineoplastic stilbene. Subsequent conversion to a useful prodrug was accomplished by phosphorylation employing in situ formation of dibenzylchlorophosphite followed 30 by cleavage of the benzyl ester protective groups with bromotrimethylsilane to afford phosphoric acid intermediate 11. The latter was immediately treated with sodium methoxide to complete a practical route to the disodium phosphate prodrug (2a). The phosphoric acid precursor (11) of phosphate 2a was employed in a parallel series of reactions to produce a selection of metal and ammonium cation prodrug candidates. Each of the phosphate salts (2a-q) was evaluated with respect to relative solubility behavior, cancer cell growth inhibition, and antimicrobial activity.
Dans l'invention, la synthèse originale de combretastatine A-2 (1a) est modifiée pour permettre d'obtenir une procédure efficace mise à l'échelle permettant l'obtention de ce stilbène antinéoplastique. Une conversion ultérieure en un promédicament utile est accomplie par phosphorilation au moyen d'une formation <i>in situ</i> de dibenzylchlorophosphite suivie (30) par un clivage des groupes protecteurs d'ester de benzyle avec du bromotriméthylsilane, ce qui permet d'obtenir un intermédiaire d'acide phosphorique (11). Ce dernier est immédiatement traité au méthoxyde de sodium pour parachever une voie pratique menant au promédicament de phosphate de disodium (2a). Ce précurseur d'acide phosphorique (11) de phosphate (2a) est employé dans une série parallèle de réactions pour produire une sélection de candidats de promédicaments cationiques d'amonium et métalliques. Chacun des sels de phosphate (2a-q) est évalué par rapport à un comportement de solubilité relative, à une inhibition de croissance de cellules cancéreuses et à une activité antimicrobienne.
