A tailor-assembly approach is employed for synthesis of complex carbohydrates wherein a polysaccharide is degraded and the shorter product obtained from the degradation is subjected to enzymatic modification to add a sugar moiety. The products may be useful in the preparation of a cancer vaccine. In one example, oligosaccharides of the type Ia group B Streptococcus (GBSIa) capsular polysaccharide and multivalent sialyl Lex antigens are specifically described. GBSIa polysaccharide was depolymerized by partial Smith degradation to fragments representing asialo core repeating units. Enzymatic sialylation of these oligomers furnished GBSIa repeating units (from monomer to pentamer). Fucosylation on GlcNAc residues of GBSIa oligomers afforded oligosaccharides that carry multiple sialyl Lex epitopes.
On met en application une approche adaptée afin d'effectuer la synthèse de glucides complexes, ce qui consiste à dégrader un polysaccharide et à soumettre à une modification enzymatique le produit le plus court obtenu à partir de cette dégradation, afin d'effectuer l'apport d'une fraction de sucre. Ces produits peuvent être utiles pour préparer un vaccin contre le cancer. Un exemple décrit de façon spécifique des oligosaccharides du polysaccharide capsulaire de Streptococcus de groupe B type Ia (GBSIa) et des antigènes de Le?X¿ de sialyle multivalents. On a dépolymérisé le polysaccharide (GBSIa) par dégradation partielle de Smith en fragments représentant des unités répétées de noyaux asialo. La sialylation enzymatique de ces oligomères a permis d'obtenir des unités répétées de GBSIa (de monomères à pentamères). La fucosylation de résidus de GicNAc d'oligomères de GBSIa a permis d'obtenir des oligosaccharides portant des déterminants antigéniques multiples de Le?x¿ de sialyle.
