The invention relates to the synthesis of polyoxygenated nitrogen systems, comprising reactions between enamines of 1,3-dioxan-5-ones and nitroolefins. More specifically, the invention relates to the reaction between enamines having formula II which are derived from 1,3-dioxan-5-ones I and nitroolefins having formula III. In this way, novel polyoxygenated nitrogen systems are generated having formula IV and V, which, as they are or following simple transformations, constitute synthetic intermediates and/or analogues of different systems of proven biological/pharmacological interest, such as certain antibiotics, or tetrodotoxin, pancratistatin or the analogues thereof. The aforementioned reactions involve the use of enamines II in Michael-type addition reactions and in direct annealing processes with double acceptor systems at relative positions 1, 3. Said acceptors include nitroolefins III, wherein R9 = CHO (alphanitroenals). The invention also relates to the preparation thereof, which is based on the oxidation of the corresponding precursor alcohols VII, and the use of same in annealing processes.
L'invention concerne la synthèse de systèmes azotés polyoxygénés moyennant des réactions entre des énamines de 1,3-dioxan-5-ones et des nitrooléfines. La présente invention concerne plus précisément la réaction entre des énamines représentées par la formule générale (II) dérivées de 1,3-dioxan-5-ones (I) et des nitrooléfines représentées par la formule générale (III). La présente invention permet d'obtenir des systèmes azotés polyoxygénés représentés par les formules générales (IV) et (V) qui, dans cette configuration ou après quelques transformations simples, constituent des produits intermédiaires de synthèse et/ou des produits analogues de divers systèmes présentant un intérêt sur le plan biologique/pharmacologique, notamment pour certains antibiotiques, ou la tétrodotoxine et la pancrtistatine et leurs produits analogues. Les réactions font intervenir les énamines (II) dans des réactions d'addition de type Michaël et des procédés d'annelage direct avec des systèmes doublement accepteurs à des positions relatives 1,3. Parmi ces accepteurs, ceux qui ressortent sont les nitrooléfines (III) dans lesquelles R9=CHO (alpha-azotés). L'invention concerne également la préparation de ces nitrooléfines, basée sur l'oxydation des alcools précurseurs (VIII) correspondants, et leur participation dans des procédés d'annelage.
