Template-fixed .beta.-hairpin loop mimetics comprising a template corresponding to one of the structures (a), (b), (c), (d), (e), (f), (g), (h) and a template-fixed chain of 4 to 20 .alpha.-amino acid residues which, if their .alpha.-C atom is asymmetric, have L-configuration can be manufactured by a novel process which is based on a mixed solid- and solution phase synthetic strategy. If desired, this process can be modified to give the enantiomers of these template-fixed .beta.-hairpin loop mimetics. These enantiomers are novel compounds, and many of said template-fixed .beta.- hairpin loop mimetics themselves are also novel compounds. The template-fixed .beta.-hairpin loop mimetics and their enantiomers can mimick flat surfaces of proteins and thus be used to probe large surface protein-protein interactions. Accordingly they can serve as lead finding tools for protein targets where it is difficult to find small-molecular-weight lead compounds.
L'invention concerne des mimétiques de boucle en épingle à cheveux .beta. comprenant une matrice correspondant à une des structures (a), (b), (c), (d), (e), (f), (g), (h) et une chaîne entre 4 et 20 résidus d'acides .alpha.-aminés qui, si leur atome .alpha.-C est asymétrique et ils présentent une configuration en L, peuvent être obtenus au moyen d'un nouveau procédé se basant sur une technique synthétique employant un mélange de phase solide et de phase liquide. Le cas échéant, ce procédé peut être modifié afin d'obtenir les énantiomères des ces mimétiques de boucle en épingle à cheveux .beta. fixés sur matrice. Ces énantiomères et nombre de ces mimétiques de boucle en épingle à cheveux .beta. fixés sur matrice sont de nouveaux composés. Ces mimétiques de boucle en épingle à cheveux .beta. fixés sur matrice et leurs énantiomères peuvent imiter des surfaces plates de protéines et, par conséquent, peuvent servir à sonder une interactions protéine-protéine à grande surface. Par conséquent, ces mimétiques peuvent servir d'outils pour trouver des composés chefs de file destinés à des cibles de protéines sur lesquelles il est difficile de trouver des composés chefs de file à faible masse moléculaire.
