A bioengineered synthesis scheme for the production of vanillin from a carbon source is provided. The bioconversion methods of the present invention comprise the steps of microbe-catalyzed conversion of a carbon source to vanillic acid followed by enzyme-catalyzed reduction of the vanillic acid to produce vanillin. As shown in the synthesis scheme of Figure 2, the microbe- catalyzed conversion step of the present invention requires five enzymes which are provided by a recombinant microbe. In a preferred embodiment, the recombinant microbe is Escherichia coli designed to cause dehydration of 3- dehydroshikimic acid and regioselective methylation of the resulting protocatechuic acid. The enzyme-catalyzed reduction step of the present invention comprises the reduction of vanillic acid to vanillin by aryl- aldehyde dehydrogenase.
Méthode de synthèse mise au point par génie génétique permettant de produire de la vanilline à partir d'une source de carbone. Les procédés de bioconversion comportent les étapes suivantes : conversion catalysée par une bactérie d'une source de carbone en acide vanillique, suivie d'une réduction, enzymatiquement catalysée, de l'acide vanillique pour produire de la vanilline. Comme le montre la Figure 2, la conversion catalysée par une bactérie requiert cinq enzymes, lesquelles sont fournies par une bactérie recombinante. Dans un mode de réalisation préféré, il s'agit de l'Escherichia Coli, qui provoque la déshydratation de l'acide 3-déshydroshikimique et la méthylation stéréosélective de l'acide protocatéchique obtenu. L'étape de réduction comprend la réduction de l'acide vanillique en vanilline par une aryl-aldéhyde déshydrogénase.
