A method and kit for detecting a target nucleic acid in a sample is described. The sample to be analyzed may include a primer which hybridizes to at least a portion of the target nucleic acid, a probe having a first region which hybridizes to at least a portion of the target nucleic acid and a second region having a detectable label, a polymerase which extends the hybridized primer and an enzyme comprising exonuclease activity that can cleave the hybridized hybridization probe to thereby generate a labeled probe fragment. At least one portion of the hybridization probe hybridizes to another portion of the hybridization probe to thereby form a folded structure. The method can involve melting the sample, reducing the temperature of the sample to allow primer and probe to each hybridize to at least a portion of single stranded target nucleic acid in the sample, elongating the primer and releasing the labeled probe fragment. The sample can be contacted with a solid support comprising surface bound capture probes which hybridize to the labeled probe fragments. The label can then be detected.
L'invention concerne un procédé et un kit pour la détection d'un acide nucléique cible dans un échantillon. L'échantillon à analyser peut comprendre une amorce qui s'hybride à au moins une partie de l'acide nucléique cible, une sonde ayant une première région qui s'hybride à au moins une partie de l'acide nucléique cible et une seconde région ayant un marqueur détectable, une polymérase qui étend l'amorce hybridée et un enzyme comprenant une activité d'exonucléase qui peut cliver la sonde d'hybridation hybridée pour générer ainsi un fragment de sonde marqué. Au moins une partie de la sonde d'hybridation s'hybride à une autre partie de la sonde d'hybridation pour former ainsi une structure repliée. Le procédé peut impliquer la fusion de l'échantillon, la réduction de la température de l'échantillon pour permettre à l'amorce et à la sonde de s'hybrider chacune à au moins une partie de l'acide nucléique cible monocaténaire dans l'échantillon, l'allongement de l'amorce et la libération du fragment de sonde marqué. L'échantillon peut être mis en contact avec un support solide comprenant des sondes de capture fixées à la surface qui s'hybrident aux fragments de sonde marqués. Le marqueur peut ensuite être détecté.
