The invention encompasses systems for column-based separations, methods of packing and unpacking columns and methods of separating components of samples. In one aspect, the invention includes a method of packing and unpacking a column chamber, comprising: a) packing a matrix material (168) within a column chamber (52) to form a packed column (70); and b) after the packing, unpacking the matrix material from the column chamber without moving the column chamber. In another aspect, the invention includes a system for column-based separations, comprising: a) a fluid passageway, the fluid passageway comprising a column chamber (132) and a flow path in fluid communication with the column chamber, the flow path being obstructed by a retaining material (148) permeable to a carrier fluid and impermeable to a column matrix material suspended in the carrier fluid, the flow path extending through the column chamber and through the retaining material, the flow path being configured to form a packed column within the column chamber when a suspension of the fluid and the column matrix material is flowed along the flow path; and b) the fluid passageway extending through a valve (130) intermediate the column chamber (132) and the retaining material (148).
L'invention porte sur des systèmes de séparations à base de colonnes, sur des procédés de garnissage et de dégarnissage de colonnes et sur des procédés de séparation de composants d'échantillons. Selon une réalisation, l'invention porte sur un procédé de garnissage et de dégarnissage d'une chambre (52) de colonne, ce procédé consistant à: a) garnir un matériau (168) matriciel dans une chambre (52) de colonne de façon à obtenir une colonne garnie (70); et b) après garnissage, retirer le matériau matriciel de la chambre de la colonne sans déplacer la chambre. Selon une autre réalisation, l'invention porte sur un système de séparations à base de colonnes, ce système comprenant: a) un passage pour le fluide qui comporte une chambre (132) de colonne et une voie pour le fluide en communication avec la chambre, cette voie étant obstruée par un matériau (148) de retenue perméable à un fluide porteur et imperméable à un matériau matriciel de la colonne en suspension dans le fluide porteur, cette voie se prolongeant dans la chambre et dans le matériau de retenue et étant conçue de façon à former une colonne garnie dans la chambre lorsqu'une suspension du fluide et du matériau matriciel s'écoule dans la voie; et b) le passage pour le fluide s'étendant dans une valve (130) placée entre la chambre (132) de la colonne et le matériau (148) de retenue.
