The present invention provides a method for treating a disease or condition in a mammal associated with influx of extracellular calcium via T type calcium channels, which comprises administering to the mammal a therapeutically effective amount of a T type calcium channel inhibitor, a prodrug thereof, or a pharmaceutically acceptable salt of said inhibitor or prodrug, wherein the T type calcium channel inhibitor blocks an .alpha.1H isoform of T type calcium channels or a .delta.25 splice variant thereof. The T type calcium channel inhibitor has a structure represented by Formula (I): Formula (I), wherein R1 is selected from the group consisting of C1-C4 alkyl, hydroxy and C1-C4 alkoxy; X is selected from the group consisting of N and CH; Z is selected from the group consisting of NH, O, S and CH2; R2 is selected from the group consisting of H, halo, NH2, C1-C4 alkyl, hydroxy and C1-C4 alkoxy; and R3 is selected from the group consisting of H, halo, NH2, C1-C4 alkyl, hydroxy and C1-C4 alkoxy. In one embodiment R1 is selected from the group consisting of C1- C4 alkyl, hydroxy and C1-C4 alkoxy, X is N, Z is O or CH2, R2 is H, halo, NH2 or hydroxy and R3 is H.
L'invention concerne une méthode permettant de traiter une maladie ou une affection d'un mammifère associée à l'influx de calcium extracellulaire par l'intermédiaire des canaux calciques de type T, consistant à administrer à un mammifère une dose thérapeutiquement efficace d'un inhibiteur de canal calcique de type T, un promédicament de celui-ci, ou un sel pharmaceutiquement acceptable dudit inhibiteur ou promédicament. Cet inhibiteur de canal calcique de type T bloque l'isoforme .alpha.1H des canaux calciques de type T, ou un variant d'épissage d25 de celle-ci. L'inhibiteur de canal calcique de type T présente la structure représentée par la formule 1: (formule I), dans laquelle R1 est choisi dans le groupe constitué par alkyle C1-C4, hydroxy, et alcoxy C1-C4; X est choisi dans le groupe constitué par N et CH; Z est choisi dans le groupe constitué par NH, O, S et CH2; R2 est choisi dans le groupe constitué par H, halo, NH2, alkyle C1-C4, hydroxy et alcoxy C1-C4; et R3 est choisi dans le groupe constitué par H, halo, NH2, alkyle C1-C4, hydroxy, et alcoxy C1-C4. Dans une forme de réalisation, R1 est choisi dans le groupe constitué par alkyle C1-C4, hydroxy, et alcoxy C1-C4, X représente N, Z représente O ou CH2, R2 représente H, halo, NH2 ou hydroxy, et R3 représente H.
