This invention provides methods for obtaining targeted gene modification in vertebrate cells using parvoviral vectors, including adeno-associated virus (AAV). The parvoviral vectors used in the methods of the invention are capable of targeting a specific genetic modification to a preselected target locus in a cellular genome by homologous pairing.
L'invention concerne des méthodes qui permettent de modifier un gène de manière ciblée dans des cellules de vertébrés au moyen de vecteurs à base de parvovirus, notamment de virus associés aux adénovirus (AAV). Les vecteurs parvoviraux utilisés dans les techniques de l'invention sont capables de provoquer, par appariement homologue, une modification génétique spécifique au niveau d'un locus cible présélectionné dans un génome cellulaire.
