Methods and compositions for altering a nucleotide sequence of interest in a plant are provided. The method involves introducing a chimeric oligonucleotide into a plant cell, wherein the oligonucleotide comprises at least two intervening blocks of RNA residues with intervening DNA residues. The RNA blocks are homologous to a plant nucleotide sequence. The chimeric oligucleotide is capable of folding to form a duplex oligonucleotide. The chimeric oligonucleotide is maintained within the nucleus of the plant whereby the alteration is introduced into the target sequences of the plant genome.
L'invention concerne des procédés et des compositions permettant de modifier une séquence nucléotidique recherchée d'un végétal. Le procédé consiste à introduire un oligonucléotide chimère dans une cellule végétale. Cet oligonucléotide comprend au moins deux blocs de résidus d'ARN comprenant des résidus d'ADN intermédiaires. Les blocs d'ARN sont homologues à une séquence nucléotidique végétale. Cet oligonucléotide chimère peut se plier de manière à former un oligonucléotide bicaténaire et est retenu à l'intérieur du noyau végétal, ce qui permet d'introduire la modification dans les séquences cibles du génome végétal.
