A method of identifying a drug that inhibits the growth or replication of a cell having a mutated MAD2 gene or an analog or homolog thereof, said method comprising the steps of : (a) contacting a cell having a mutated mAD2 gene or an analog or homolog thereof with the drug; and (b) determining whether the drug modulates the activity of a wildtype secondary gene which is synthetically lethal when it is mutated and is present in combination with mutated MAD2 gene or an analog or homolog thereof. A screening assay system for identifying a drug, and a pharmaceutical composition comprising an effective amount of a drug and a pharmaceutically acceptable carrier or diluent.
L'invention concerne une composition et des procédés servant à identifier une ou plusieurs cibles secondaires de médicaments et leur utilisation dans l'identification de médicaments ou de candidats médicaments, notamment pour le traitement du cancer ou de troubles de réplication cellulaire. Selon l'invention, on utilise des cribles létaux synthétiques à base de levures pour identifier et valider fonctionnellement de nouveaux gènes cibles afin de tuer les cellules présentant des défauts dans les mécanismes de cycle cellulaire. Ces gènes cibles nouvellement identifiés peuvent servir à mettre au point de nouveaux agents thérapeutiques pour traiter le cancer, la croissance cellulaire bénigne et les infections aux levures.
