There is provided a method for preparing an orally disintegrating tablet (ODT) composition comprising microparticles of one or more taste-masked active pharmaceutical ingredients, rapidly-dispersing microgranules, and other optional, pharmaceutically acceptable excipients wherein the ODT disintegrates rapidly with saliva in the buccal cavity forming a smooth, easy-to-swallow suspension. Furthermore, the microparticles (crystals, granules, beads or pellets containing one or more actives) with a taste-masking membrane applied by a modified solvent coacervation process comprising a water-insoluble polymer and at least one gastrosoluble inorganic or organic pore-former, exhibit a pleasant taste when placed in the oral cavity and provide rapid, substantially-complete release of the dose on entry into the stomach.
Cette invention concerne un procédé permettant de préparer une composition pour des comprimés à dissolution orale (ODT) comprenant des microparticules d'un ou de plusieurs principes actifs pharmaceutiques au goût masqué, des microgranules à dissolution rapide et, éventuellement, d'autres excipients pharmaceutiquement acceptables. La composition ODT se dissous rapidement avec la salive dans la cavité buccale formant, ainsi, une suspension lisse facile à avaler. En outre, les microparticules (cristaux, granules, billes ou pastilles contenant un ou plusieurs principes actifs) dotées d'une membrane permettant le masquage du goût appliquée lors d'un procédé de coacervation de solvant modifié comprenant un polymère non soluble dans l'eau et au moins un porogène organique ou inorganique gastrosoluble, présentent un goût agréable lorsqu'elles sont placées dans la cavité buccale et elles permettent une libération rapide et essentiellement complète de la dose posologique lorsqu'elles atteignent l'estomac.
