A process for synthesizing an mRNA having a high translational template activity which comprises transferring a sequence having a specific sequence and a sequence structure having at least a non-translational sequence respectively into the 5'-end and 3'-end of an mRNA which is a protein translational template molecule to thereby construct an mRNA having a high translation efficiency, and constructing a plasmid having a wide applicability which contains a template DNA for transcribing the above mRNA; a purification method; a practically available means of cell-free synthesis of proteins by using the same wherein a continuous RNA synthesis means is combined with a continuous cell-free protein synthesis means with the use of a wheat germ extract to construct a transcriptional/translational non-coupling cell-free system for synthesizing proteins; and a method whereby a plural number of proteins can be co-expressed which is constructed by using the means of cell- free synthesis of proteins.
L'invention concerne un procédé de synthèse d'un ARN messager présentant une activité matricielle translationnelle élevée. Ce procédé consiste à transférer une séquence comprenant une séquence spécifique ainsi qu'une structure de séquence comportant au moins une séquence non translationnelle respectivement dans l'extrémité 5' et l'extrémité 3' d'un ARN messager se présentant sous la forme d'une molécule matrice translationnelle protéique, d'où la construction d'un ARN messager doté d'une efficacité translationnelle élevée, puis à construire un plasmide présentant vaste champ d'applications et contenant un ADN matrice destiné à transcrire l'ARN messager susmentionné. L'invention concerne également un procédé de purification ainsi qu'un dispositif utilisable en pratique de synthèse acellulaire de protéines faisant appel à cette molécule matrice, un dispositif de synthèse d'ARN continue étant combiné à un dispositif de synthèse acellulaire continue de protéines grâce à l'utilisation d'un extrait de germe de blé, d'où la construction d'un système acellulaire transcriptionnel/translationnel permettant de synthétiser des protéines sans couplage. L'invention se rapporte en outre à un procédé permettant la coexpression d'une pluralité de protéines, ledit procédé étant mis en oeuvre au moyen de ce dispositif de synthèse acellulaire de protéines.
