The present invention relates to methods for preparing bioscaffolds useful in the repair of tears. More specifically, the invention relates to a method of treating rotator cuff tear in a mammalian subject in need thereof comprising the steps of: (i) selectively expanding tenocytes in vitro in culture medium comprising insulin or a functional derivative and a glucocorticoid or a glucocorticoid-like molecule to produce a culture of expanded tenocytes; (ii) seeding a bioscaffold with said expanded tenocytes to produce a tenocyte seeded bioscaffold; and (iii) implanting said tenocyte seeded bioscaffold proximal to the rotator cuff tear. The present invention also relates to a bioscaffold comprising cells, wherein more than 80 % of said cells are tenocytes.
L'invention porte sur des procédés permettant de préparer des échafaudages biologiques utiles pour réparer des déchirures. L'invention concerne plus spécifiquement un procédé permettant de traiter une déchirure de la coiffe des rotateurs chez un sujet mammifère atteint, ledit procédé comprenant les étapes suivantes : (i) développement sélectif in vitro des ténocytes dans un milieu de culture comprenant de l'insuline ou un dérivé fonctionnel et un glucocorticoïde ou une molécule de type glucocorticoïde afin d'obtenir une culture de ténocytes développés; (ii) ensemencement d'un échafaudage biologique avec lesdits ténocytes développés afin d'obtenir un échafaudage biologique ensemencé avec un ténocyte; et (iii) implantation dudit échafaudage biologique ensemencé avec un ténocyte à proximité de la déchirure de la coiffe des rotateurs. La présente invention concerne également un échafaudage biologique incluant des cellules, plus de 80% desdites cellules étant des ténocytes.
