A method of detecting transmissible spongiform encephalopathies is described which involves (a) treating a sample of tissue, blood or a blood derivative from a test subject with alcohol and a detergent, (b) adding an agent which degrades normal prion protein (c) adding an agent which denatures abnormal prion protein, (d) adding a prion-specific antibody and (e) detecting binding of the antibody to the sample. Also described is a method of fixing abnormal prion protein to a substrate comprising (a) treating the protein or a sample suspected to contain the abnormal prion protein with alcohol and an anionic detergent and (b) adding a protease, in the presence of the substrate.
La présente invention concerne une méthode permettant de détecter des encéphalopathies spongiformes transmissibles. Ladite méthode consiste à (a) traiter un échantillon tissulaire, sanguin ou un dérivé sanguin d'un sujet à tester avec de l'alcool et un détersif, (b) ajouter un agent qui dégrade une protéine prion normale (c) ajouter un agent qui dénature une protéine prion anormale, (d) ajouter un anticorps spécifique du prion et (e) détecter la fixation de l'anticorps sur l'échantillon. L'invention concerne également une méthode de fixation d'une protéine prion anormale sur un substrat consistant à : (a) traiter la protéine ou un échantillon supposé contenir la protéine prion anormale avec de l'alcool et un détersif anionique et (b) ajouter une protéase, en présence du substrat.
