Hyaluronidase activity in human semen is found to be related to sperm concentration, motility, morphology, and the rate of fertilization in vitro. A method for assessing sperm fertilizing ability is provided. In the method, semen samples are treated with detergent to extract hyaluronidase and applied to a mixture of hyaluronic acid and agar. Enzyme activity is assessed by measuring the area of substrate hydrolysis. Hyaluronidase activity is found to be significantly related to the success of fertilization and to the fertilization rate. These results indicate that measurement of hyaluronidase activity can provide a useful method for assessing the integrity of the acrosomal enzyme system, providing a simple, reliable, and cost effective predictor of the fertilizing potential of human sperm.
