There is provided a method for the synthesis of norbuprenorphine, and ultimately buprenorphine, utilizing oripavine as the starting material. Conventional methods of producing buprenorphine utilize thebaine as the starting material, requiring an O-demethylation step, typically a low to moderate yield transformation. The present use of oripavine as a starting material does not require an O-demethylation step, since the oripavine molecule lacks an O-3 methyl group.

La présente invention a pour objet une méthode de synthèse de la norbuprénorphine, puis de la buprénorphine, ladite méthode utilisant l~oripavine au titre de produit de départ. Les méthodes classiques de synthèse de la buprénorphine utilisent la thébaïne au titre de produit de départ, ce qui nécessite une étape de O-déméthylation, transformation qui est généralement de rendement faible à moyen. L'utilisation de l~oripavine selon la présente invention au titre de produit de départ ne nécessite pas d'étape de O-déméthylation, dans la mesure où la molécule d~oripavine ne comprend pas de groupement O-3-méthyle.
