The invention is based on observing that LIF is capable of activating the self-renewal of tumor stem cells in cancer, in particular gliomas, which indicates that the inhibition of LIF, and generally of IL- 6 type cytokines, can be used in therapeutic compositions for the treatment of diseases associated with unwanted proliferation. The invention also relates to a method for the identification of compounds capable of blocking/inhibiting the proliferation of stem cells, and to an in vitro method for the diagnosis of diseases associated with unwanted cell proliferation in a subject or for determining the predisposition of a subject to suffer said disease associated with unwanted cell proliferation, or for prognosis of average life expectancy of subjects suffering from said tumors.
L'invention est basée sur l'observation selon laquelle LIF peut activer l'auto-renouvellement de cellules souches tumorales dans le cancer, en particulier, les gliomes, ce qui indique que l'inhibition de LIF, et généralement des cytokines de type IL-6 peut être utilisée dans des compositions thérapeutiques pour le traitement de maladies associées à une prolifération indésirable. L'invention concerne également un procédé pour l'identification de composés capables de bloquer/inhiber la prolifération de cellules souches et un procédé in vitro de diagnostic de maladies associées à une prolifération cellulaire indésirable chez un sujet ou pour déterminer la prédisposition d'un sujet à souffrir de ladite maladie associée à une prolifération cellulaire indésirable, ou pour le pronostic de l'espérance de vie moyenne de sujets souffrant desdites tumeurs.
