Thermal Denaturation Screening Assay to Identify Candidate Compounds for Prevention and Treatment of Parkin-son's Disease The invention provides highthroughput screening assays to identify agents useful for treatment of Parkinson's Dis-ease. In one embodiment the assay includes exposing a plurality of test samples, each containing a test compound and parkin pro-tein, to thermal destabilization conditions and determining parkin ligase activity in the test samples relative to a control sample not containing a test agent. A test agent contained in a test sample in which parkin ligase activity exceeds the ligase activity in said control sample is identified as a candidate compound for treatment of Parkinson's Disease.
L'invention porte sur des analyses de criblage de haut débit pour identifier des agents utiles pour le traitement de la maladie de Parkinson. Dans un mode de réalisation, l'analyse comprend l'exposition d'une pluralité d'échantillons d'essai, chacun contenant un composé d'essai et la protéine parkin, à des conditions de déstabilisation thermique, et la détermination de l'activité de la ligase parkin dans les échantillons d'essai par rapport à un échantillon témoin ne contenant pas d'agent d'essai. Un agent d'essai contenu dans un échantillon d'essai dans lequel l'activité de la ligase parkin dépasse l'activité de la ligase dans ledit échantillon témoin est identifié comme étant un composé candidat pour le traitement de la maladie de Parkinson.
