The mesophilic restriction endonuclease AvaI and its isoschizomers derived from Anabaena and Anabaenopsis have been found to support highly efficient amplification of target nucleic acid sequences in thermophilic Strand Displacement Amplification (tSDA). The maximum amplification factors observed occurred at temperatures which were previously reported to result in reduced enzyme activity. As AvaI is an isoschizomer of BsoBI (a thermophilic restriction endonuclease commonly used in tSDA), the invention provides an alternative enzyme which is more readily available than BsoBI and does not require the redesign of SDA amplification primers previously constructed for use in the BsoBI system.

On a découvert que la restriction mésophile endonucléase AvaI et ses isoschizomères dérivés d'Anabaena et d'Anabaenopsis aidaient l'amplification hautement efficace de séquences d'acide nucléique ciblé dans le déplacement amplifié du brin thermophile (tSDA). Les facteurs d'amplification maximums observés ont été atteints à des températures qui avaient auparavant été notées pour réduire l'activité de l'enzyme. AvaI étant un isoschizomère de BsoBI (un endonucléase thermophile de restriction communément utilisé dans le tSDA), l'invention offre un enzyme de rechange qui est plus facilement disponible que BsoBI et n'exige pas la modification d'amorces d'amplification de SDA déjà construites pour utiliser dans le système BsoBI.
